He was a Member of Parliament in the National Assembly for the Moshupa-Manyana constituency from 2009 to 2018.

Masisi’s early political journey makes an interesting read. He had served under former Minister, late Lesego Motsumi who was a Specially Elected MP , and at the Office of the President. When Khama redeployed Motsumi to serve in the Foreign Service in India for alleged unsatisfactory performance, Masisi took over as the substantive Minister in the Presidency. He continued to cement his relationship with Khama and became one of the most visible Ministers.

Earlier in his life, say 1980s, Masisi became a high school social studies teacher after graduating from the University of Botswana in 1984 in English and History. He taught at Mmanaana Secondary School in 1984 in Moshupa village before moving on to the University of Botswana in 1987 as a curriculum development specialist.

In 1989, he went to Florida State University to obtain a Master's degree in social sciences education, after Masisi met some FSU faculty members working in Botswana for the Junior Secondary Education Improvement Project. Following graduation, UNICEF in Botswana employed him.
